The Arignar Anna Bus Stand is located along Palayamkottai Road, which stretches westward and connects to the main commercial areas of the city.

Thoothukudi Railway Station is one of the oldest railway stations in India. The station was declared a Model Station in 2007, and infrastructure developments continue at the site. There is also a second station at Tuti-Melur with suburban connections.

The daily Pearl City Express service connects Thoothukudi with Chennai, departing at 7:35 PM in the evening and arriving the following morning at 7:30 AM. Night train services also operate from Bangalore and Mysore, departing at 9:20 PM and 6:00 PM respectively and arriving at 10:45 AM the next day. Regional destinations with direct train services include Coimbatore and Tirunelveli.
